In the DELTARUNE Chapter 5 PC Version 0.0.0241 changelog, it is stated that "The last boss of the chapter will now get gradually easier after losing an excessive amount of times in one play session." How many times do you have to lose for the last boss of the chapter to get easier, and does "one play session" mean it won't get easier if I quit the game and reopen?

According to the Deltarune wiki, the boss will start getting easier after 6 deaths.
Yes, restarting the game is considered a new "play session" and will reset the difficulty back to normal. You can also quit to title and re-load your save for the same result.
The wiki also lists the exact difficulty changes:

I also just tested it myself to confirm and quitting to title and then loading the save back up will reset the difficulty adjustment.
