Dana Buckman reposted thisDana Buckman reposted thisHeavy metals — cadmium, chromium, mercury, lead — are persistent, non-degradable contaminants of the food chain. EFSA's figures place dietary cadmium exposure in European adults at the boundary of the Tolerable Weekly Intake. The gut is where ingested metals become systemic burden. We assessed three single-strain lactobacilli — Lactiplantibacillus plantarum LP14, Lactobacillus crispatus LCR04, Lactobacillus acidophilus LA12 — along a multilevel ladder: static in vitro, dynamic SHIME, gut-ex-vivo at CAAD-UPO. Viable cell concentration was tracked compartment by compartment by flow cytometry in AFU; ICP-MS partitioned each metal between supernatant and cell pellet, the resolution required to distinguish sequestration from matrix artefact. Three findings emerged. In the colonic phase of the SHIME, LCR04 displaced ~45 percentage points of lead from supernatant into pellet relative to blank, 20–40 for cadmium and chromium with LCR04 and LP14, ~10 for mercury. Lead, refractory in static broth, became reachable the moment the strain was given a colonic environment. LA12, the strongest static performer, failed to proliferate in the colon and removed no metal. Sequestration tracked metabolic activity, not biomass: only strains expanding by ~2 log AFU — LP14 and LCR04 — achieved meaningful removal. In the gut-ex-vivo on mouse small intestine, the metal mixture compromised barrier integrity: paracellular permeability rose, claudins and occludin dysregulated, TNF-α and IFN-γ elevated, IL-10 suppressed. Pre-incubation with any of the three cell-free supernatants restored the phenotype: permeability to baseline, transcripts in range, cytokines toward homeostasis. Sadly, heavy-metal contamination is a chronic-exposure problem the regulatory machinery cannot eliminate at source. What our data support is a deliberately narrow proposition. A properly selected single-strain probiotic, validated along a transit-realistic ladder, could operate as a gut-level adjunct to existing food-safety measures. Not a substitute for source control. Not a clinical treatment. An adjunct — a further layer of protection at the interface where ingested metals become systemic burden. This paper, just out in Frontiers in Microbiology, is my PhD thesis — Cycle XXXVI of the Food, Health and Longevity programme at Università del Piemonte Orientale. Open access. DOI in first comment. #probiotics #flowcytometry #shime #publichealth #heavymetals #foodsafety #microbiome

Dana Buckman shared thisThis is fantastic to see. Solid clinical validation is exactly what the biotic space needs right now to differentiate and drive credibility. Kudos to the Probiotical team for investing in the science that moves the whole industry forward. 🔬✨Dana Buckman shared this“Clinically studied” on a supplement label can mean almost anything — from a single pilot in healthy volunteers to a multi-year programme across independent research groups. The difference is not semantic. It is the distance between a data point and a body of evidence. Over the past ten years, we have built one of the most comprehensive clinical programmes in psychobiotics around a single four-strain formula: L. fermentum LF16, L. rhamnosus LR06, L. plantarum LP01, B. longum 04. Four billion live cells per day. Eight peer-reviewed publications. Four independent trials: Italy, US, Australia. Marotta et al. (2019) established proof of concept in 38 healthy adults: depression scores down 44%, anger down 37%, sleep quality improved 29% — all persisting three weeks after washout. It was also the first study with published stability data reported in AFU and enumerated by Flow Cytometry. Calgaro et al. (2021) confirmed strain recovery and three baseline bacteriotypes predicting response. Maltodextrin placebo showed a bifidogenic effect — sharpening every subsequent study. Walden et al. (2023) scaled to 70 subjects in the largest dedicated psychobiotic RCT. Anxiety and rumination decreased significantly versus placebo. Plasma serotonin rose at six and nine weeks — no change in placebo. Kerksick et al. (2024), on the same cohort, confirmed significant improvements in sleep quality, latency, and disturbance (PSQI). Di Cesare et al. (2023) added the metabolomic layer: 600 MHz NMR, 840 urine samples, 80% classification accuracy. The discriminating metabolites — hippurate, hydroxyphenylacetate, formate, amino acid shifts — all bacteria-derived. Moschonis et al. (2026): a clinical population with subthreshold depression, DSM-5 diagnosed (conducted during COVID!). Questionnaires replaced by blood and saliva. Fasting glucose decreased with a significant treatment-by-time interaction. hs-CRP decreased. Glutathione — the body’s primary endogenous antioxidant — increased at both six and twelve weeks. Cortisol decreased significantly. Dacaya et al. (2025) documented the SSRI-like serotonin regulation — the biphasic rise-then-fall mirroring antidepressant pharmacodynamics. The follow-up (Moschonis et al., 2026) expanded the picture. LBP and sCD14 — markers of endotoxin translocation — decreased significantly, indicating barrier strengthening. GIP, an enteroendocrine hormone, also decreased, suggesting metabolic benefits beyond mood. We named this findings BIFIZEN! Five layers: microbiome, metabolome, serotonin, inflammation, clinical outcomes. The most significant findings — bacteriotype response, biphasic serotonin, barrier strengthening — were not in the original hypotheses. Eight publications. Three countries. Ten years. That is what “clinically studied” looks like when you mean it :) Full biblio in the first comment. #psychobiotics #probiotics #gutbrainaxis #clinicaltrials #microbiome #stress #anxiety #sleep #BIFIZEN

Dana Buckman shared thisJoin me in beautiful Vancouver for the upcoming IPA + Probiota Americas meeting! The momentum in our industry right now is incredible. I’m excited to look at where this fast-moving space is headed—let's connect while we're there!Dana Buckman shared thisMost viability measurements tell you what already happened. What if they could help guide manufacturing decisions in real time instead? At Probiota Events 2026 in Vancouver, our CEO at BioForm Solutions, Dana Buckman, and Dr. Rishi R. Trivedi, PhD from Biose Industrie will be sharing a collaborative poster focused on applying flow cytometry as a real-time process tool for next-generation probiotic manufacturing. For highly sensitive anaerobic organisms like Akkermansia muciniphila, traditional plating methods can create long feedback loops that are difficult to use during active manufacturing. This work explores how faster physiological insights can support better decision-making across fermentation, downstream processing, formulation, and stability monitoring. As the NGP space continues to evolve, the industry needs analytical approaches that move beyond simple enumeration toward a deeper understanding of microbial health and product robustness throughout the manufacturing lifecycle. If you’ll be at Probiota, Dana would love to connect and continue the discussion around the future of real-time viability monitoring in microbiome therapeutics. #NGP #microbialhealth #probiotics #microbiome https://lnkd.in/gYPMsKP4

Dana Buckman reposted thisDana Buckman reposted thisWe don’t have a probiotic demand problem. We have a validation problem. The team here at BioForm Solutions came across this recent piece on why probiotics often don’t work and it reinforces something many of us in the space have been seeing firsthand. From strains that aren’t actually effective, to viability issues, to the simple reality that some microbiomes won’t even accept new bacteria - the variability is real. And yet… the category continues to grow. That’s the tension right now. The next phase of this industry won’t be driven by more SKUs or better marketing. It’s going to be driven by precision, measurement, and proof - understanding not just what’s in the product, but what’s alive, functional, and actually doing something meaningful. Because if we can’t consistently answer that, the rest doesn’t matter. Curious how others are thinking about this - are we finally at the point where validation becomes the differentiator? https://lnkd.in/grqf4PJF

Dana Buckman liked thisDana Buckman liked thisEvery time I see a probiotic label that lists "𝘓𝘢𝘤𝘵𝘪𝘱𝘭𝘢𝘯𝘵𝘪𝘣𝘢𝘤𝘪𝘭𝘭𝘶𝘴 𝘱𝘭𝘢𝘯𝘵𝘢𝘳𝘶𝘮" and stops there… (Yes, the strain is the entire point. The species is just the postcode.) In probiotics, the strain is the grape, the vintage and the estate. A wine list that just says "red" is not a wine list. It is a guess. The species level on a probiotic label is the same guess in a lab coat. Footnote: Two strains of 𝘓𝘢𝘤𝘵𝘪𝘱𝘭𝘢𝘯𝘵𝘪𝘣𝘢𝘤𝘪𝘭𝘭𝘶𝘴 𝘱𝘭𝘢𝘯𝘵𝘢𝘳𝘶𝘮 can sit in completely different evidence bases, with completely different GI, immune or metabolic dossiers behind them. The DSM, CECT, ATCC or NCIMB code in tiny grey font on the back of the box is doing all the scientific work. The big number on the front of the box, the lonely species name, the green leaf icon are doing none of it. If the strain code is missing, the trial is missing.

San Diego's hottest new fast food spot...is for cells? 🦠 Local biotech company Nucleus Biologics is helping cell and gene therapy companies get their cell food faster thanks to its "cellular Nespresso machine". With a push of a pod, labs and clinics can have 500 liters of cell food in 3 hours—less than half the time it would traditionally take to whip up. It's a shift that will help cut R&D costs down, so more companies can afford to keep working on life-saving cell and gene therapies.

Developing new regenerative medicine treatments is only the first step; ensuring that Californians have access to these treatments is just as important. To put clinical trials and therapies within reach of Californians, CIRM has approved an Access & Affordability strategy that outlines how CIRM will deliver cell and gene therapies to Californians. Many patients, especially those in underserved or remote areas, face barriers to care and affordability. This strategy focuses on improving accessibility at clinical sites, collaborating with payers to support favorable policies, and planning early in research to reduce patient costs. CIRM's Patient Support Program is also expanding with a $15.6 million Patient Assistance Fund to help cover costs for patients participating in California-based clinical trials. Maria Gonzalez Bonneville, Chair of the Access & Affordability Working Group, said, "A patient's home address and income should not determine their treatment options. CIRM is committed to expanding equitable access to clinical trials, prioritizing underserved communities statewide." Read full release: https://bit.ly/4sgVAWP
