Our portfolio company Complement Therapeutics announced the completion of Cohort 1 dosing in the Phase I/II Opti-GAIN study evaluating CTx001 in Geographic Atrophy (GA) secondary to AMD, alongside a favourable recommendation from the Independent Data Monitoring Committee (IDMC) to proceed with dose escalation into Cohort 2. CTx001 is an investigational AAV gene therapy designed to downregulate multiple complement pathways through sustained local expression following a single subretinal injection, with the potential to offer a differentiated approach to treating GA. The Opti-GAIN study is also among the first in GA to prospectively evaluate ellipsoid zone (EZ) integrity and focal microperimetry as structural and functional endpoints, aiming to provide a more sensitive assessment of disease progression and potential treatment effect at the individual patient level. This milestone marks an important step forward for Complement Therapeutics and highlights continued innovation in the development of next-generation therapies for complement-mediated diseases and vision loss. Forbion portfolio company Prilenia has announced a new Data Use Agreement with CHDI Foundation to advance Huntington’s disease research through secure sharing of placebo-arm clinical data from the PRIDE and PROOF studies. The collaboration aims to improve understanding of placebo effects and strengthen future clinical trial design in HD. By enabling broader access to high-quality clinical datasets, Prilenia is helping accelerate innovation and research efficiency across the field. Our portfolio company Mestag Therapeutics announced the first patient has been dosed in the Phase I STARLYS trial evaluating MST-0312 in patients with advanced solid tumors. MST-0312 is a FAP-targeted bispecific antibody designed to activate LTBR in the tumor microenvironment and induce tertiary lymphoid structures (TLS) and high endothelial venules (HEV), with the goal of enhancing anti-tumor immune responses. The STARLYS study will evaluate MST-0312 as both a monotherapy and in combination with Keytruda® (pembrolizumab), assessing safety, pharmacokinetics, pharmacodynamics, and early anti-tumor activity across selected solid tumors. This milestone marks an important step forward for Mestag’s fibroblast immunology platform and its approach to addressing tumors with limited response to current immunotherapies. We are encouraged by the positive Phase 2 data reported by our portfolio company Enterprise Therapeutics for ETD001, its investigational inhaled ENaC blocker for the treatment of cystic fibrosis (CF). The study achieved its primary efficacy endpoint, demonstrating a clinically relevant and statistically significant improvement in lung function versus placebo in people with CF who do not benefit from CFTR modulators, a patient population representing approximately 10% of individuals living with CF and among those with the highest unmet medical need.
